Abstract

Objective To explore the mechanism of atorvastatin (AVT) on inhibition of invasion in glioma U87 cells.Methods To detect the efficiency of AVT on inhibition of the invassion of glioma cells by using Transwell kit,RT-PCR and Western blot methods were used to study the effects of AVT on CXCL12 and CXCR4 expression.Results The results showed that the effect of atorvastatin (AVT),after 24 hour treatment,is an dose-dependent decrease in CXCR4mRNA expression with the increasing concentration of atorvastatin.Compared with control group,the cells treated with 1.0,2.0,8.0 μmol/ L AVT have showed statistically significant changes (P values are 0.045,0.021,0.002 respectively).At the same concentration of AVT (2.0 μmol/ L),the expression level of CXCR4 mRNA decreased,following a time-dpendent manner (12,24,36 hours),and being statistically significant (P values are 0.040,0.019,0.001 respectively).The AVT administration does not have effects on the expression of CXCL12 mRNA,statistically,with P value being 0.869.Conclusion AVT significantly inhibited the invasion of glioma U87 cells clinically,and inhibited the expression of CXCR4 experimentally.Therefore we speculate that the AVT maybe inhibit glioma cell invasiveness through downregulation of CXCR4 expression.
