Abstract

The behavior of a variety of alkylsilyl derivatives of adenosine were studied on GLC using silicone gum rubber as the liquid phase. A re-examination of the retention time of newly prepared cytidine showed it to be longer than the other major nucleosides using gas-liquid chromatographic methods. Although evidence for a silyl derivative of cytidine monophosphate was obtained using paper chromatography, it could not be detected with GLC. Infra-red, ultra-violet, and paper chromatography also indicated a silyl derivative of nicotinamide adenine dinucleotide, but only nicotinamide mononucleotide and adenosine monophosphate were found to pass through GLC. Silylated polyadenylic acid hydrolyzates were successfully chromatographed by GLC. The findings were discussed with reference to transfer ribonucleic acid analysis.
