Abstract

Large amounts of SV40 virus were obtained using a SV40 large-plaque (LP) mutant in a high passage line of BS-C-1 cells. A procedure of concentration and purification without the use of sonication of treatment with trypsin and deoxycholate is described. The diameter of the full virus particles in three-dimensional crystals was found to be not larger than 41.1 ± 0.6 mμ. The partial specific volume of the full virus particles was v = 0.658 ± 0.0004 / Full virus particles had a sedimentation constant of 210 S and a diffusion coefficinet of 0.90 × 10 −7 cm 2/second. The molecular weight of the particle was calculated to be 17.3 × 10 6.
