Abstract

The need for and value of a goniometer stage and stereo electron microscopy to obtain maximum information from preshadowed carbon replicas of frozen biological specimens has already been documented. Now it is important that one be forewarned of the possibility of misinterpretation of the morphology of an object if only a single electron micrograph or a single stereo pair is used for interpretation.Surface replicas of the outer surface of frozen begonia petals were prepared in the usual manner. The large pieces of cleaned replica were lifted onto uncoated 75 mesh grids and examined in the JEM 100-B electron microscope with goniometer stage.
