Abstract

1. 1. A new culture system (adapted from the procedure of Seglen, 1975) of 9-week-old chicken hepatocytes is presented, validated by a hepatic enzyme activity time-course (stearyl-CoA desaturase) during a 6-day period. 2. 2. Stearyl-CoA desaturase activity was measured in isolated and cultured hepatocytes from genetically lean or fat chickens. A significantly higher activity was shown in the isolated hepatocytes from the fat line as compared to the cells of the lean line. This difference disappeared after 24 hr of culture. 3. 3. The triglyceride secretion by cultured hepatocytes was determined and a correlation with the animal adiposity was shown. 4. 4. These results obtained on a hepatocyte culture system directly confirm previous observations showing that adiposity would be related to VLDL processing and secretion, and monoene availability in the chicken hepatocytes.
