Abstract

Potato cv. `Red Pontiac was cultured and multiplied through several passages in vitro. The microplants and mini-tubers produced from two-year old in vitro cultures were investigated for phenotypic stability under glasshouse and plastic tunnel. Only one branch of a single plant among the many thousand derived from tissue culture showed variegated leaves. However, a few off-type mini-tubers were observed for changed tuber skin colour, which propagated true to type in the subsequent propagation. Tuber skin colour was markedly affected with the planting season. Red skin tubers planted in late autumn produced light pink tubers under unheated plastic tunnel during off-season from September to December. However, tuber shape, skin colour and skin texture were stable traits, and could be used for detecting off-types among mini-tubers derived from tissue cultured plants.
