Abstract

Carnitine is a quaternary ammonium compound which is naturally synthesized in the body. But depending upon diet, age, and other factors, supplementation of l-Carnitine may be needed to maintain adequate levels in the body. l-Carnitine is highly moisture-sensitive, which makes it difficult to stabilize in various dosage forms. In the current research work, primarily stability enhancement process was done to overcome stability problems due to the hygroscopic nature of l-Carnitine through the pro-drug approach. The resulting pro-drug compound showed excellent stability in terms of moisture absorption. The percentage of moisture uptake of drugs has been reduced from 42.5% to less than 3%. This also gives the acceptable organoleptic property in contrast with l-Carnitine. Stability enhanced l-Carnitine was formulated as fast dissolving pellets using various super disintegrates by extrusion/spheronization technique. The drug-loaded pellets exhibited very less disintegration time and excellent drug release in 5 min.
