Abstract

Despite expressing high levels of Polycomb group proteins, embryonic stem cells (ESCs) are refractory toH3K27 trimethylation, notably at super-enhancers regulating key pluripotency genes. In this issue of Molecular Cell, Wang etal. (2017) report that the histone chaperone Spt6 prevents H3K27 trimethylation of key ESC super-enhancers.
