Abstract
The spectrofluorodensitometric assay for the determination of flurazepam and its major metabolites in blood involves selective extraction of flurazepam and its major metabolites into ether and hydrolysis in acid to their respective benzophenones, which are cyclized in dimethylformamide-potassium carbonate to their respective 9-acridanone derivatives and separated by TLC. The fluorescence of the 9-acridanones is measured directly on the chromatoplate, using a scanning TLC analyzer in the reflectance mode. The limits of detection of the assay are 0.5-1.0 ng of each compound/ml of blood using a 4-ml specimen/analysis.
