Abstract

Neuronal noradrenaline is stored in two types of organelles: large and small dense cored vesicles (1, 2). Large vesicle preparations of high purity have been obtained from bovine splenic nerve, while hitherto only heavily contaminated fractions of small vesicles have been obtained from vas deferens and heart. We have improved the isolation of small vesicles by using the vas deferens of the castrated rat, as castration leads to an initial muscle atrophy and therefore a relative increase in neuronal material. The vesicle preparation, consisting of 25–30% small vesicles has been characterized biochemically and morphologically (in detail described by us elsewhere 3), and was compared to large noradrenergic vesicles.
