Abstract

Magnetic Tweezers (MT) provide a powerful tool to study the relaxation and gate dynamics of DNA topoisomerases by enabling the manipulation and study of changing DNA topology in real time. DNA topoisomerases regulate DNA supercoiling and relieve any structural tangle that happens during normal cellular processes such as replication and transcription where the topoisomerase undergoes conformational changes. We use a duplex DNA construct with a ssDNA gap of 27 nt with a topoisomerase recognition site in the gap to observe the gate dynamics of wild type and mutant Mycobacterium smegmatis topoisomerase I (MsmTOP1).
