Abstract
It only takes one. Polyketide biosynthesis on modular polyketide synthases (PKSs) depends on the specific interaction between multiple protein subunits. These recognition events are mediated in part by docking domains at the termini of the multienzymes. Site-directed mutagenesis within a model PKS system shows that even single amino acid substitutions within the docking domains can significantly alter the efficiency of docking.
