Abstract
A method is described for the simultaneous determination of protease inhibitors Indinavir, Saquinavir, and Ritonavir in human plasma using liquid chromatography with ultraviolet and electrochemical detection. Sample pre-treatment consisted of solid-phase extraction prior to ion-pair reversed phase high performance liquid chromatography with ultraviolet detection at 240 nm (Saquinavir and Ritonavir) and electrochemical detection at + 750 mV (Indinavir and Saquinavir). The method was validated with respect to specificity, linearity, limits of detection and quantification, precision, and accuracy. The proposed LC assay was utilized to analyze plasma samples from patients treated with HIV-protease inhibitors.
