Abstract

The analytical ion-exchange method for the rapid and simple separation of free nucleoside mono-, di and triphosphates in the tissue was presented by high-pressure liquid chromatography. Free nucleotides were extracted from tissue by a simplified method using a methylal-ethanol mixture. The simultaneous analysis of free nucleotides can be carried out in less than 40 minutes on the column of LiChrosorb-NH2 with a linear gradient of potassium phosphate buffer. In the profile of nucleoside monophosphates, IMP and XMP which had been difficult to separate from other nucleotides were identified satisfactory by a single column chromatography. Our chromatographic method was applied to the simultaneous analysis of free nucleotides prepared from guinea pig epidermis and liver.
