Abstract

A simple smear technique for the sampling and culturing of embryonal and fetal cells with the aid of monofil meshes is described. Skin smears provided successful cultures in 5/6 and 8/8 embryonal smears and appeared to be more suitable than the smears from the surface of amnion and umbilical cord. These cultures, except those derived from the umbilical cord, retained long-term proliferation activity in EPL medium enriched by Eagle's MEM vitamins without calf serum as did the cultures of amniotic fluid cells. The present method offers direct and gentle sampling of cell material for prenatal genetic diagnostic at cell level, even in the early phases of development.
