Abstract

Objective To study the significance and protein expression of tumor suppressor gene PLAGL1 in humor hepatocellular carcinoma (HCC) and mRNA expression in L02 and HepG2 cells.Methods Thirty cases of HCC tissues and 30 cases of tumor-adjacent tissues were collected from patients surgically treated in our hospital.The protein expression of PLAGL1 was detected by immunohistochemistry.The normal liver cell line L02 and hepatoma cell line HepG2 cells were cultured,and the expression of PLAGL1 mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR).Results PLAGL1 protein expression in tumor-adjacent tissues ( n = 27,90% ) was significantly stronger than in HCC tissues (n = 2,7% ) (χ2 = 38.44,P < 0.05 ).RT-PCR results showed that PLAGL1 mRNA in the L02 cells was significantly higher than in HepG2 cells.Conclusion Down-regulation of PLAGL1 protein may be correlated to the occurrence and development of HCC.PLAGL1 protein expression was paralleled with the PLAGL1 mRNA down-regulation.
