Abstract
1. 1. Optimum conditions have been determined for the measurement of serum lipase with respect to emulsifying agent, buffer, pH of hydrolyzate, and time of hydrolysis. 2. 2. The procedure developed makes use of the first hour of hydrolysis of triglyceride. 3. 3. Normal human serum lipase values range from 2 to 12 units per milliliter. A unit is the amount of enzyme that catalyzes the liberation of 1 μmole of fatty acid from triolein or olive oil in 1 hr per milliliter of serum. 4. 4. In acute pancreatitis markedly elevated serum lipase values were observed when determined with olive oil as substrate.
