Abstract

To the Editor: Hemoglobin (Hb) is the major erythrocyte protein, and measurement of free Hb in serum can be used as a marker of hemolysis. Classically, cyanohemoglobin spectrometry and immunonephelometric methods have been used for assaying free Hb in serum (1)(2). Hb metabolism partly occurs in tissue macrophages, which can engulf senescent erythrocytes (extravascular hemolysis) or take up Hb released from ruptured erythrocytes (intravascular hemolysis) and immature erythrocytes in the bone marrow. Because of the oxidative and toxic properties of the iron-containing heme in Hb, Hb is complexed by the plasma protein haptoglobin (Hp).
