Abstract

Blocking of 125I-fibronectin (Fn) binding to two protein-A-defi-cient mutants U-320 and U-305 derived from Staphylococcus aureus SA113(83A) was tested, using various human and murine sera. Normal human sera completely inhibited Fn binding to both the mutants. However, sera obtained from patients suffering from acute staphylococcal infections, caused a high degree of inhibition of Fn binding only to U-320, whereas the binding to U-305 was less affected. Also, murine antisera against whole cells of Staphylococcus aureus laboratory strains, Cowan 1 and Newman, or to cloned Fn binding protein (FnBP) blocked the binding of 125I-Fn to U-320 but not to U-305. The Fn binding components of the two mutants showed marked differences in the susceptibility to proteolytic digestion. Our data suggest the existence of more than one type of FnBP in Staphylococcus aureus. The difference in the blocking of 125I-Fn binding to mutants U-320 and U-305 suggests antigenic variation among staphylococcal FnBP.
