Abstract

Objective To screen the microRNAs (miRNA, miR) fingerprint of peripheral blood in patients with recurrent gastric cancer by Luminex technique. Methods 100 patients with gastric cancer recurrence, 100 patients with non-recurrence and 100 heathy controls were recruited in this study. Five kinds of miRNAs in peripheral blood were screened by Luminex technique. In addition, 50 patients with gastric cancer were collected and miRNAs expression in serum was detected dynamicly to select microRNAs fingerprint in serum of patients with recurrent gastric cancer. Results The expression of five kinds of miRNAs in preoperative serum of patients with gastric cancer was significantly higher than in the healthy controls [The median expression in gastric cancer and healthy controls was as follow: for miR-10b (8.25 vs. 3.25); for miR-21 (11.18 vs. 3.25); for miR-183 (22.86 vs. 4.13); for miR-371-5p (13.57 vs. 3.37); and for miR-378 (18.19 vs. 3.99), all P=0.000). Postoperative serum miR-10b in patients with early gastric cancer recurrence was not significantly upregulated as compared with that in the patients with early gastric cancer non-recurrence (median: 7.41 vs. 7.33, P=0.336). The expression of miR-21, miR-371-5p, miR-378 and miR-183 in 50 patients with gastric cancer was significantly downregulated after operation and significantly upregulated when recurrence. The miR-183 was removed from fingerprint due to miR-183 was not significantly upregulated when recurrence (The median: 1.99 vs. 1.14). Conclusion The three miRNAs (miR-21, miR-371-5p and miR-378) were screened as the specific miRNAs diagnostic fingerprint of gastric cancer recurrence and could predict early recurrence of gastric cancer.
