Abstract

AbstractAlthough affinity chromatography is an efficient separation method, impediments to its large scale applications include difficulty in predicting column behavior after scaling‐up and lack of suitable solid matrix support for fast flow operations. The introduction of radial‐flow chromatography and a modified cellulose support has overcome the impediments. Radial chromatography has advantages in low pressure drop at fast flow rate and in linear scaling‐up. The performances of the cellulose support is studied by attaching two bioaffine ligands to it and observing the efficiency in enzyme purification. Application of a radial‐flow affinity cartridge to remove proteases in human plasma shows 70% efficiency after one‐pass treatment.
