Abstract

Laser Raman spectroscopy has many useful applications in the study of biological macromolecules including proteins, peptides, nucleic acids, carbohydrates, and lipids. In addition to regular and resonance Raman important information concerning the molecule's conformation, stability, and side chain environments can be revealed by studying the heat perturbation and denaturation of proteins and peptides. It is therefore necessary to devise an apparatus which can heat a sample to a precise temperature and maintain that temperature while a spectrum is recorded. In this note we describe an apparatus for thermostating samples contained in capillary tubes. The heating block was designed for use with a Spex Ramalog 5 spectrometer but could be adapted to any other type of spectrometer.
