Abstract

Transient receptor potential canonical (TRPC) channels are non-selective, calcium-permeable cation channels. Previously, it has been demonstrated that calcium ions bind directly to the S2-S3 linker of TRPC4 channels. However, the effect of direct calcium binding on gating of TRPC4 channel has not been identified. Candidate residues E417, Q420, N435, D438 in the S2-S3 linker were selected based on structure, and in this study, we elucidate that S2-S3 linker is crucial for gating of TRPC4 channel in a calcium-dependent way.
