biopsies by real-time PCR. Additionally, 6 healthy controls were analyzed. Results: mRNA and protein expression of TKTL1 and DNase X were found in all carcinoma cell lines but in none of the CEC cultures from healthy controls. Both TKTL1 and DNase X expression were independent from culture conditions such as cell density, culture time and spheroid size. In the murine colon carcinoma model, biopsies of affected areas showed significantly more DNase X expression as compared to unaffected areas (603±147 versus 208±183, p<0.01). In addition, there was a significant difference between biopsies of DSS/azoxymethane colon cancer mice and the 6 healthy control (603±147 versus 135±198, p<0.02) mice. In contrast, the expression of DNase 1 was comparable in both affected and unaffected areas of colon cancer mice and healthy control mice. Conclusion: Our data demonstrate that TKTL1 and DNase X expression is induced in human and murine colon cancer cells. TKTL1 and DNase X may serve as targets for novel cancer diagnostics.
