Abstract

The vascular corrosion cast with resin MercoxΤΜ-CL-2B for visualization under scanning electron microscopy (SEM) it allows the analysis of the angioarchitecture. For in such a way it has necessity of partial or total maceration of the organic tissue around of the mold. With relation to the corrosion, each organ to be analyzed presents individual techniques, entailed to the size and the type of the tissue that the specimen forms. In the rats tongues it has necessity to eliminate two layers in the dorsal surface, before displaying the vascular cast with resin. Few standardized protocols with relation to the maceration of tongues exist. Of this form, 9 male rats Wistar had been used, adults, with 2 months of age and approximately 200g of weight, manipulated under intraperitonealy anesthesia for accomplishment of the technique of vascular corrosion cast. From there, 3 tongues had been macerated with concentrations of NaOH the 1, 10 and 20%, respectively, with standardized temperature in 45 oC, for 10, 24 and 36 hours. All the gotten molds from now on, had been submitted to the drying for the point-critical and placed in metallic stubs, for sputtering with two carbon layers, followed for a gold layer. When used solutions of 10 and 20% for maceration, it was possible to surprise the formation of structures that make it difficult this process, as well as deposits on the specimens, of form that these are extremely dirty. The period between 24 and 36 hours is necessary for removal of the keratin of the dorsal surface of the language of rats with solution of sodium hydroxide 1% in the conditions of this study.
