Abstract

Objective To investigate the expression of RNASET2 in pancreatic cancer and its significance.Methods The expression levels of RNASET2 mRNA and in pancreatic cancer (32 cases),adjacent benign tissue (24 cases),3 human pancreatic cancer cell lines and benign ductal epithelial cell line were quantitated by using reverse transcriptase-polymerase chain reaction (RT-PCR).The protein expression of RNASET2 was evaluated by Western blotting in 8 fresh frozen tissues and immunohistochemistry (IHC) in 10 paraffin-embedded specimens.Results RNASET2 was down-regulated significantly in pancreatic cancer (6.516 ±3.033,P <0.05) and cell lines BxPC-3,PANC-1 and SW1990 (2.814 ±0.325,2.342 ±0.454 and 5.473 ±0.682,P <0.01) respectively.The protein expression of RNASET2 was obviously down-regulated in pancreatic cancer tissues and tumor cell lines.The group with perineural invasion showed lower expression of RNASET2 (5.021 ± 3.102,P < 0.05).Conclusion The expression of RNA-SET2 was down-regulated in pancreatic ductal adenocarcinoma and tumor cell lines,and this gene might be correlated to tumorigenesis and perineural invasion of pancreatic cancer.
