Role of Macrophages in Immunity and Pathogenesis of Experimental Cryptococcosis

Abstract

Summary Alveolar and peritoneal macrophages isolated from mice during different stages of cryptococcal infection were able to phagocytize Cryptococcus neoformans. Alveolar macrophages exhibited a fungistatic effect on Cr. neoformans at day 14 post infection, whereas peritoneal macrophages displayed a fungicidal effect on the organism at days 21 and 28 post respiratory exposure to Cr. neoformans. However, these effects were transient and both macrophage populations provided a favorable environment for the intracellular growth of Cr. neoformans as a result of “an immunosuppressive state” which occurred during progressive stages of cryptococcal infection. Therefore, rather than being the main effector arm of acquired immunity against progressive cryptococcosis, the macrophages may contribute to the spread of Cr. neoformans by acting as a mediator for the dissemination of the organism to the central nervous system. Zusammenfassung Während untershiedlimer Stadien der Cryptococcrrs-Infektion wurden von Mäusen alveolare und peritoneale Makrophagen isoliert, die Cryptococcus neoformans-Zellen phagozitieren konnten. Die alveolaren Makrophagen wiesen am 14. Tag nach der Infektion einen fungistatischen Effekt gegen Cr. neoformans auf, während die peritonealen Makrophagen diesen Effekt am 21. und 28. Tag n a h der aerogenen Infektion mit Cr. neoformins zeigten. Dieser fungistatische Effekt war jedoch zeitlich begrenzt, und beide Makrophagentypen erwiesen sich als eine günstige Umgebung für das. intrazelluläre Wachstum von Cr. neoformans. Dies ist möglicherwaise die Folge eines „immunsuppressiven Zustandes”, zu dem es im Laufe der progressiven Stadien der Cr. neoformans-Infektion kam. Der Makrophage ist demnach nicht der überzeugend wichtigste Faktor für die erlangte Immunität gegen die progressive Cryptokokkose, er kann vielmehr die Streuung des Pilzes in das Zentralnervensystem, in dem er als Überträger wirkt, fördern.
