Abstract

The study aimed to investigate E. coli in the surfaces and pulp of gallstones by isolating and diagnosing them from gallstones and identifying most types of stones that form inside the biliary sac. The study included 40 patients with cholecystitis, the samples were collected from different age groups (10-80) years, (6) males and (34) females, who underwent surgery at Al-Hussein Teaching Hospital for the period from October 2020 to February 2021. The stones are removed from the gallbladder and then washed with Normal saline, the stones samples are the external surface and the inner core , each placed in the nutrient medium such as brain heart Infusion broth (BHI) for 24 hours at a temperature of 37 ° C in order to stimulate the potential presence of bacteria, then transferred to the culture medium that selective for E.coli ( chromogenic agar) for a period of 24 to 48 hours in order to diagnose and isolate the bacteria and assure it with biochemical examinations . The media used are UTI chromogenic agar, E. coli forms chromogenic agar. The study explained that there are different types of gallstones and according to the percentages of their presence in men and females, where cholesterol stones are most common types that reached from the masses of 21 samples in females 90.47% and in men 9.52%. Pigment stones reached from a total of 10 samples were 80 % for females, 20 % for men. Mixed stones totaled 7 samples for both sexes in females 57.14% and men 42.85%. Our results also found that the number of females who were recorded to have gallstones was higher compared to men. The age group most affected by cholelithiasis, and for both sexes, was of age 41-50 are (68.75%) for females and (31.25%) for men. while they are very few within 10-20 years, (100 %) in females and (zero) in men. The results of the transplantation for biliary sac stones were given positive bacterial culture for cholesterol stones with a higher percentage than the pigment stones. Gallstones were cultured (surface and pulp) where the Escherichia coli bacteria gave great positive results for the cultivation compared to the rest of the bacteria, where was the percentage of presence E .coli on the surfaces (71.05%) and pulp (55.26%) of stones for different types of gallstones. Gallstones are a common disease at different ages, where females are affected at higher rates compared to men, there are three types of gallstones: cholesterol, pigment and mixed. E.coli may be considered one of the causes of gallstones by giving it positive results of cultured on the surfaces and pulp of the gallstones.
