Abstract

Objective To study the expression of Dectin-1 in bladder cancer and its roles in bacillus calmette-guerin (BCG) intravesical therapy.Methods Immunohistochemistry and Western blotting were used to detect the expression of Dectin-1 in bladder cancer tissue as well as T24,J82 and EJ cell lines.These cells were stimulated with different concentrations (0,0.1,1.0,2.0,10.0,20.0 g/L) of BCG for 15,30 min,1,2,4 and 8 h,and the nuclear factor-κB (NF-κB) pathway activation was detected before and after blocking Dectin-1 receptor.The levels of cytokines were determined by enzyme linked immunosorbent assay (ELISA).Results Dectin-1 was expressed in the surface of bladder cancer cells.Stimulation with 1.0 g/L of BCG for 1 h could significantly activate NF-κB signaling in T24,and the high-est secretion levels of interleukin (IL)-6 and tumor necrosis factor (TNF)-α were found in this concentra-tion group,which were separately up to (29.10 ± 2.89) and (21.57 ± 2.88) ng/L,respectively.The BCG stimulating effect could be weakened when Dectin-1 receptor was blocked.Conclusion BCG could activate NF-κB signaling pathway and inflammatory immune response through Dectin-1 receptor,which may play an important role in BCG intravesical therapy.
