Abstract Specific peptide bond formation with the α-amino group of lysine requires prior blockade of the ε-amino group. The necessary differentiation can be achieved by using the fact that the side-chain amino group is the more basic and nucleophilic because there is an electron-withdrawing group next to the α-amino group (Scheme 6.1), or by taking advantage of the fact that the two functionalities can engage in chelate formation (Scheme 6.2).
