Abstract

A Talking Point contribution1 dealing with a topic as controversial as the role of ceramide in apoptosis inevitably provokes an animated discussion. We are delighted to see that in their letter2, Perry and Hannun have addressed the reliability of the diglyceride kinase (DGK) assay for ceramide quantification. Their conclusion – that this assay is prone to artifacts if lyophilized preparations of DGK or low amounts of enzyme are used – underscores our suggestion that ceramide data based on this method need a careful re-examination. Now that alternative assay methods are available and that the dangers of the DGK method have been addressed2,3, we are confident that future research on this topic will proceed with due caution.
