Abstract

Objective To study the relationship between polymorphism of monocyte chemoattractant protein(MCP)-1 and kidney failure in elderly patients with type 2 diabetes mellitus( T2DM ). Methods The polymorphism was detected with polymerase chain reaction-restriction fragment length polymorphism (PCRRFLP). We investigated -2518A/G polymorphism of MCP-1 gene in elderly patients with T2DM and kidney failure (n = 56) ,compared with matched elderly T2DM patients without nephropathy (diabetic controls, n=56) and healthy controls (n = 50). Results The overall genotypic distribution of -2518A/G in MCP-1gene has no difference in patients with T2DM compared to healthy controls. Although the genotype has no significantly difference between patients with T2DM and kidney failure and the diabetic controls, the A allele was more frequent in patients with T2DM and kidney failure than that in diabetic controls ( P < 0.05 ). The carriage of A allele was significantly associated with kidney failure. Conclusion The -2518A allele of MCP-1 gene is associated with kidney failure in elderly patients with T2DM. Key words: Kidney failure; Type 2 diabetes mellitus; MCP-1 gene; Gene polymorphism
