Abstract

The manXYZ (ptsL, pel/ptsM) locus consists of 3 genes involved in mannose transport in Escherichia coli K12. In addition, the manY gene product is required for penetration of λDNA into the cell. We have previously constructed a series of lacZ gene fusions to manYZ. In this paper we describe the molecular cloning and regulatory properties of these fusions. A λ transducing phage was isolated that carried both the manX gene and the manYZ-lacZ gene fusion. The manX gene together with the fusion was subcloned into the fusion cloning vector pMLB524. Analysis of these subclones showed that lacZ was fused to manY. The manX and manY genes appeared to be in the same operon, manX being the promoter-proximal gene.
