Abstract

Objective To investigate the effects of salvia miltiorrhiza on gene expression of interstitial collagenase and type I collagen in HSC-T6 cell. Methods Hepatic stellate cell ( HSC-T6 ) was cultured in medium containing different concentration of salvia mihiorrhiza (0. 8, 0. 4 g/L) and cells were collected after" incubation for 48 hours. Total RNA of HSC was isolated and gene expression levels of interstitial collagenase (MMP13) and type I c?llagen were measured by reverse transcription polymerase chain reaction. Results MMP13 gene expression level of different groups of salvia mihiorrhiza (0. 8, 0. 4 g/L) were 0. 25 + 0. 05, 0. 27 ± 0. 06 respectively and that of control group was 0. 26 ± 0. 04. Type I collagen gene expression level of different groups of salvia mihiorrhiza (0. 8, 0. 4 g/L) were 0. 76 +0. 10, 1.55± 0.23 respectively and that of control group was 3.12 ±0.46. Conclusion Salvia miltiorrhiza may significantly inhibit type I collagen gene expression of HSC-T6 and the effects were correlate with the dosage. Sal- via mihiorrhiza has no effect on MMP13 gene expression. Key words: Salvia mihiorrhiza; HSC-T6 cell; Interstitial collagenase ; Type I collagen ; Gene expression
