Abstract
Objective To investigate the effects of carthamus tinctorius on gene expression of stromelysin-1 (MMP3) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in HSC-T6 cells.Methods HSC-T6 cells were cultured in the medium containing different concentrations of carthamus tinctorius (1.0,and 0.5 g/L) and collected after incubation for 48 h.Total RNA of HSC-T6 cells was isolated and gene expression levels of MMP3 and TIMP-1 were measured by using reverse transcription polymerase chain reaction (RT-PCR).Results MMP3 gene expression levels in 1.0 and 0.5 g/L carthamus tinctorius groups were 2.80 ±0.36 and 2.52 ±0.32 respectively,and that in control group was 2.13 ±0.30.TIMP-1 gene expression level in 1.0 and 0.5 g/L carthamus tinctorius groups were 1.66 ± 0.23 and 2.04 ± 0.30 respectively,and that in control group was 4.03 ± 0.56.Conclusion Carthamus tinctorius may enhance the expression of MMP3,but significantly inhibit TIMP-1 gene expression of HSC-T6 cells in a concentration-dependent manner.
