Abstract

Assays of enzyme activity in fractions obtained after Percoll density gradient certrifugation of protoplast extracts from Mesembryanthemum crystallinum performing exclusively C3 photosynthesis or Crassulacean acid metabolism (CAM) established the cytoplasmic location of phosphoenolpyruvate (PEP) carboxylase and NADP-malic enzyme, the mitochondrial location of NAD-malic enzyme, and the chloroplastic location of pyruvate, Pi dikinase. The activity of these enzymes was elevated in the CAM tissue, and net synthesis of PEP carboxylase protein increased during CAM induction. This compartmentation and regulation of enzymes of the carbon pathway implicate the transport of pyruvate and PEP across the chloroplast envelope during the metabolism of malate in the light in the CAM tissue.
