Abstract
Abstract Chain recombination studies were carried out with two purified “monofocal” anti-DNP antibody preparations and purified serum anti-DNP antibody. The results indicated that after separation and recombination of H and L chains the “monofocal” antibodies completely regained their hapten-binding capacity whereas serum antibody showed a 10-fold decrease in the association constant of its binding with hapten. The regain in activity of “monofocal” antibodies is attributed to their homogeneity, and the loss of activity upon separating and recombinating chains of serum antibody is the result of chain mispairing rather than of denaturation. These results serve as further evidence for the homogeneous nature of “monofocal” antibodies and their probable monoclonal origin.
