Abstract
Biochemical and morphological modifications occurring in the pancreas of Bio Breeding (BB) pre-diabetic rats and Wistar non-diabetic rats subjected to an immunosuppressive treatment with Cyclosporin A (CyA; 10 mg/kg body wt daily/4 weeks, administered in lipid soluble form interperitoneally) were observed: (i) directly after treatment withdrawal, and (ii) 30 days after the end of the treatment. Biochemical assays showed that CyA significantly enhances glycaemia ( P<0.00001) in all treated groups. Insulin levels were not significantly altered in both the Wistar groups but showed a significant decrease in both BB rat groups when measured at the end of treatment ( P<0.001). This decrease continued in the BB rats tested 30 (days after the end of treatment ( P<0.0001). Morphological findings (light microscopy and transmission electron microscopy) demonstrated the presence of vacuolated areas in the B cell cytoplasm of the pancreas belonging to animals observed directly after treatment withdrawal. Thirty days after the end of treatment the morphology returned to the normal appearance in nearly all ( n=9/10) of the Wistar rats and in only 6/10 of the BB rats.
