Abstract

Background: Patients with diabetes have a high incidence of peripheral vascular disease and limb loss despite vascular reconstruction. These experiments were designed to evaluate the temporal progression of acute limb ischemia reperfusion injury in a murine model of type 2 diabetes (Db, Leptin receptor mutant). Methods: Db and the respective wild type (Wt) mice were both subjected to 3 hours of ischemia followed by either 1 (1DR) or 7 (7DR) days of reperfusion. At the end of each experimental interval, mice were sacrificed, and their limbs were assayed for mitochondrial activity (MA, an index of tissue viability, expressed as percent contralateral limb).
