Abstract
The modern concept of enamel structure depicts a highly mineralized tissue with an organic matrix, of which part is assumed to consist of fibres of a “keratin-like” protein. It is generally believed that the fibres of the matrix are formed before the deposition of mineral and that this functions as a framework for the mineral crystallites. It is further assumed that the presence of fibres is necessary for the nucleation and orientation of apatite crystallites. Careful study of amelogenesis in mammals, using electron microscopy has failed to reveal the fibrous structural elements in exactly the form predicted by current concepts of enamel formation. It is suggested, therefore, that the concept of a “ fibrous” organic matrix of enamel be re-examined.
