Abstract

A new capillary electrophoresis method was developed for the separation of four phenylpropanoid glycosides (teucrioside, verbascoside, teucrioside-3””-O-methyl ether and teucrioside-3””, 4””-O-dimethyl ether) from T. chamaedrys. The electrophoretic separation was performed using a 50 cm × 50 μm i.d. (41.5 cm effective length) fused silica capillary. A phosphate-borate buffer (pH 8.9) in 15% (v/v) isopropanol was used as running electrolyte. The samples were injected by pressure for 3 seconds at 50 mbar and the running voltage was 25 kV at the injector end of the capillary. The separation of four phenylpropanoid glycosides could be achieved within 10 min with good repeatability. The method was validated in terms of reproducibility, linearity, accuracy and applied for the quantitative determination of teucrioside, teucrioside-3””, 4””-O-dimethyl ether and verbascoside. This is the first analytical method developed for the analysis of four phenylpropanoid glycosides from T. chamaedrys by capillary electrophoresis. The influence of pH, organic modifiers and composition of the electrolyte on resolution were investigated. Other Teucrium species were analyzed.
