Abstract
The effects of TDZ (N-phenyl-N′-1,2,3-thidiazol-5-ylurea) and CPPU (N-(2-chloro-4-pyridil)-N′-phenylurea) on bud initiation of Amaranthus edulis thin cell layers (tTCLs) explants were investigated. tTCLs (0.2–0.4 mm thick) excised from 1 week old seedlings were cultured on Murashige and Skoog (MS) basal salts medium supplemented with varying concentrations of TDZ or CPPU ranging from 1 to 20 μM and 1 week after culture, buds were initiated. The optimal concentrations were 2 μM for TDZ and 10 μM for CPPU. Buds developed into normal plants after transfer onto a MS basal salts medium containing BA to obtain normal internode elongation and onto a MS basal salts medium containing IBA or TDZ for rooting.
