Abstract

Objective To rapidly identify wild strains and vaccine strains of varicella zoster virus(VZV) by PCR-RFLP in Jiangxi province in 2010-2015. Methods We have collected 112 herpes fluid and throat swab specimens from clinical diagnosis cases of varicella zoster in Nanchang from 2010 to 2015, extracted virus DNA from the specimens, used the specific primers of ORF62, ORF38 and ORF54 areas, and used restriction enzymes SmaⅠ, PstⅠ and BglⅠto do fragment length polymorphism analysis. Results We have analysised results the of 39 positive PCR products and all the products can be digested by SmaⅠ-, PstⅠ+ and BglⅠ+ , but the RFLP result of vaccine is SmaⅠ+ , PstⅠ- and BglⅠ+ . Conclusions The wild strains and vaccine strain of VZV be identified by the method of PCR-RFLP and the VZV the specimens collected in Jiangxi Province from 2010 to 2015 are all wild strains.
