Abstract

In order to improve accuracy and speed of recombinant human Antithrombin III (rhAT III) analysis, a rapid method for quantitative determination of rhAT III during cell culture processes and subsequent purification steps was developed. The developed procedure is based on reversed phase high performance liquid chromatography (RP-HPLC) using a C4 column. The separation by gradient elution with water and acetonitrile takes less than 20 minutes even if crude samples, such as serum containing cell culture supernatant, have to be analyzed. Automation and a high sample throughput are possible with this reliable method. Furthermore, insulin, transferrin and albumin can simultanesously be quantified with minor changes of the elution gradient.
