Abstract

Ozonated oils had an increasing interest lately due to the importance of these products in medicine. Therefore, plenty of studies are needed to conduct. In this paper Quantitative and qualitative analysis of ozonated olive oil samples was applied as well as stability studies were done during storage of those samples on shelf. For qualitative studies, H-NMR spectroscopy was applied. For quantitative determination, ozone efficiency method was used to determine the percentage of ozone in those oils depending on the calculating of peroxide value, and the method was validated. In order to monitor the stability of the ozonated olive oil, ozone efficiency method was used on the samples and it was supported by Acidity Value method. In H-NMR spectroscopy, the samples were dissolved in deuterated chloroform CDCl 3 then the analysis was accomplished using Bruker Avance 500 spectrometer. In ozone efficiency method, Ozonated oil samples were dissolved in a mixture of chloroform and glacial acetic acid then, the whole mixture was titrated with sodium thiosulfate solution after adding of saturated potassium iodide solution to calculate peroxide value of the sample then its ozone efficiency. In validation of the proposed method linearity, accuracy, and precision were evaluated according to ICH guidelines. Stability study was done using OE method in particular points and was continued for six months taking into account the temperature of storage. The results of H-NMR spectra showed that by ozonation of olive oil some peaks was disappeared related to double bonds. At the same time, new peaks were emerged indicating to ozonides formation. Moving to ozone efficiency method validation results which showed that the proposed method gave accepted linearity (R 2 = 0.9984) with range of weights (0.2-0.4g), with excellent mean recovery values (98-102%), and precision with RSD% less than 2%. OE, PV, and AV increased faster during the storage at high temperature. In conclusion, H-NMR is a reliable and effective method for qualitative determination of ozonated oils. Moreover, ozone efficiency method is an effective method in determining ozone percentage in ozonated oils as well as its simplicity and capability to use in quality control laboratories.to maintain ozonated olive oil stable the temperature should be taken into account as well as the initial value of the measurements.
