Abstract

A sensitive and precise quantitative densitometric HPTLC method has been established for analysis of palmitoyl hexapeptide both as the bulk drug and in formulations,. Silica gel 60 F254 plates were used with toluene-ethanol 9:1 as mobile phase. This system gave compact spots for palmitoyl hexapeptide (RF 0.33 ± 0.02). Densitometric quantification was performed in absorbance mode at 221 nm and well resolved bands were obtained. Regression analysis of the calibration data revealed a good linear relationship (mean value of r2 = 0.99910 ± 0.003) between response and amount of palmitoyl hexapeptide in the range 10–30 ng per band. The mean slope and intercept were 114.5 ± 0.002 and 311.1 ± 1.32, respectively. The method was validated for precision, recovery, robustness, ruggedness, and specificity. The limits of detection and quantification were 3 and 9 ng per band, respectively. The method is selective and specific with potential application in cosmetics and pharmaceutical analysis.
