Abstract

ABSTRACT A quantitative gas chromatographic method for the routine determination of urinary 11-deoxy-17-ketosteroids as their trimethylsilyl ethers, and of pregnanediol as the diacetate on a single column with 1 % SE-30 and 2% XE-60 as stationary phase, has been described. After hydrolysis, the 17-ketosteroids are purified by alumina column chromatography, converted into the trimethylsilyl ether derivatives, and quantitated by gas-liquid chromatography. Two procedures are described for the analysis of pregnanediol, using alumina column chromatography or Girard T separation as preliminary purification. Final purification is achieved by thin-layer chromatography (TLC) of the pregnanediol diacetate, followed by gas-liquid chromatography (GLC). The method has been applied to study the urinary excretion of androsterone, etiocholanolone and dehydroepiandrosterone in normal subjects before and after administration of corticotropin, Metopirone and dexamethasone. ACTH administration caused a marked increase in the...
