Purification and Properties of the Nitrile Hydratase of a New Strain of Rhodococcus sp.

Abstract

A strain of bacteria with nitrile hydratase activity was isolated from soil. Taxonomic studies indicated that the strain 7 belongs to the genus Rhodococcus. Its nitrile hydratase was purified and crystallized (from the crude extract). The enzyme has a molecular mass of about 278,000 Da. and consists of two kinds of subunits, with molecular masses of 26,000 Da. and 32,000 Da. The enzyme was active toward various aliphatic and aromatic nitriles. Ein Bakterienstamm mit Nitril-Hydratase-Aktivität konnte aus Erde isoliert werden. Taxonomische Studien ordnen den Stamm 7 der Gattung Rhodococcus zu. Die Nitril-Hydratase wurde gereinigt und kristallisiert (aus Rohextrakt). Das Enzym hat ein Molekulargewicht von 278.000 Da. und besteht aus zwei Arten von Untereinheiten mit Molekulargewichten von 26.000 Da und 32.000 Da. Das Enzym hydrolysiert verschiedene aliphatische und aromatische Nitrile.
