Abstract

This study focused on the cell behaviour study by culturing HeLa cell on several simple protein tests on the substrate with the assist of high voltage pulsed electric field inducement towards wound healing studies. Cell behaviour changes can be seen when various test applied to it. Which cell behaviour can lead to several of result such as cell attachment, guidance, migration, and proliferation which influenced the growth of cells, however, it depends on the test and parameter applied to the cell. This study uses high voltage pulsed electric field with a parameter of 2kV voltage, 30µs pulse duration and single pulse. HeLa cells were cultured on several substrates. Firstly, the cell was plated on the free surface, then tested on self-assembled monolayer and free surface, and lastly, the cell was tested on the micropatterned surface by using the microcontact printing method. All this test were done with the assist of high voltage pulse electric field inducement. The finding shows a positive response of cell with the pulse electric field inducement by showing an increment of cell growth compared to control, while the protein deposition test had shown that it influenced cell to move and migrate towards protein side, and the grid of protein influenced cell to attach and follow the shape of the protein patterned surface. These positive responses will ultimately contribute towards wound healing studies.
