Abstract

Objective Employing proteomics approaches to screen the proteins related to human idiopathic interstitial pneumonia (IIP) and to gain theoretical data for illuminating the pathogenesy and prognosis of IIP. Methods Fresh lung tissues of local anesthesia small cut open lung biopsy (8 cases) and normal lung tissues away from benign lung tumors (5 cases) were collected. Total proteins were extracted and separated by two-dimensional gel electrophoresis (2-DE). Different expression proteins were analyzed using Imagemaster 2D 5.0 image analysis software. Different proteins were identified using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. Biological information of these proteins were searched in database of these peptide mass finger-printing. Western blotting and RT-PCR were performed to examine the expression of Annexin. Results Establishing two-dimensional electrophoresis profiles, 7 differential protein spots were found. The high abundance protein spots were analyzed and identified, which were heat shock protein 70, Annexin Ⅱ , Haptoglobin and so on. Western blotting and RT-PCR confirmed expression of AnnexinⅡ increased in IIP compared with normal controls. Conclusions Well-reproducible 2-DE maps of IIP patients and normal controls are established and some proteins functions are identified. AnnexinⅡ may play important roles in IIP pathogengsis, and it may participate through mechanisms of fibrolysis system disequilibrium. These results will be useful to find out some proteins related to pathogenesis of IIP and will provide scienticfic foundation for screening molecular biomarker used to diagnose and treat IIP.
